How to initiate the Leica TCS SP2
control panel
- Switch buttons from right to left (PC-Scanner-Laser)
- Before switching on the argon laser, check that the power Level is set to MIN. Once the laser is switched on you, can turn the power up (labeled mark).
- Argon laser for laser lines: 458, 477, 488, 514. To switch on first turn the key to START, it then jumps automatically back to the ON position.
- Switch on 543nm and 633nm laser line if needed.
software initiation
- Make sure that the upper stage limit is set before starting the Leica Confocal Software.
- Choose “Personal” and start the initialization process.
basic acquisition setup
- In the Aquire toolbar, select beam to open the Beam Path Setting window.
- There you can set up all the aquisition parameters according to your sample and needs.
- Use the control panel to adjust for image gain and offset
NOTE : This is a rather manual set up! YOU HAVE TO ...
- switch between scanning and visible mode ...
- change filter wheels (GFP, RFP filter for observation by eye, empty position for scanning) ...
- change objectives ...
... BY HAND!!!
