Data from fluorescence imaging offers the benefit of both spatial and temporal resolution as well as multi-parametric analysis of cells in heterogenous populations. This technical seminar will provide an introduction to fluorescence, discussion of the properties of fluorescent dyes and proteins. Strategies for engineering fluorophores to optimize them for imaging applications will be described using recently developed technologies to illustrate these approaches. Considerations for labeling live and fixed cells will be presented with an emphasis on super-resolution microscopy. |

September 12, 2014: Chemical identification and surface measurements on nanoparticles using the NanoTweezer | Seminar: Optofluidics - NanoTweezer Technology | In this talk, Dr. Abbey Weith overview Optofluidics’ NanoTweezer
Technology, the first commercial system capable of performing chemical identification and direct surface measurements on single nanoparticles in solution. Using proprietary nanophotonic devices and microfluidics, the NanoTweezer works by directly optically trapping small scale objects in solution and analyzing their scattered light, enabling the system to extract meaningful physical and chemical parameters. Unlike traditional free space optical traps which are light diffraction limited and thus can only influence dielectric objects larger than a hundred nanometers in size, the NanoTweezer technology is powered by proprietary nanophotonic waveguides which are engineered to exert stronger optical forces via light confinement and resonance, enabling a technological leap in trapping and analysis capability. The talk will cover the technology’s working principles (including waveguide capturing and single particle Raman spectroscopy) and its applications to nanomedicine, nanotoxicology and other fields. The system is commercially available in Germany. |

Light-sheet microscopy is an emerging technology that enables imaging of biological specimens with minimal photo-damage. It opens new avenues to study cell biological and developmental processes with unprecedented imaging speed. It also facilitates systems biology approaches by quantitative imaging of entire living systems. Light-sheet microscopy has only recently left the optical tables of technology developers and became more broadly available through commercial and open-access platforms. This labeling step can perturb the specimen and is often not desired e.g. in investigations in vivo. Vibrational spectroscopy, such as Raman and infrared spectroscopy, is a label free technique that provides direct molecular and structural information. The spectra of these complementary methods are hence referred to as molecular fingerprint. Infrared spectroscopy imaging and Coherent anti Stokes Raman spectroscopy (CARS) are relatively new techniques for a very fast and sensitive label-free microscopy of cells and tissues. We will present applications of these fast, label-free and in vivo techniques, among others for studying spinal cord regeneration in several animal models (rat, axolotl) and for observing tumor cells both in vivo and in cell culture. |

March 4, 2014: Refractometer Demo | DEMO | We have the pleasure to invite you all for the demonstration of an advanced refractometer, the J457 from Rudolf Research Analytical, which might well suit the needs of the community of microscopists of the whole campus. Indeed, this machine has among all the other characteristics a measurement range of refractive index that goes from 1.26 to 1.72, it's highly accurate, temperature control (10-110°C), and can measure through solid matter too (e.g. animal tissues). Seminar: 11:15 MPI-CBG Galleria Practical Part: Available for testing for one week in MPI-CBG LMF wet lab (room 036) It is very important for us all to understand the real capabilities of the machine. And since Mr. Schmidt, who will come for the demonstration, would be available to test real samples the on 4th of March, please bring your media, specimens, and challenging tissues. In the case you would like to measure solid materials, just consider that the diameter of the sample should be less than 10 mm, and that hard samples like glass have to be polished. |

Built around a Nikon TiE stand it will feature two Yokogawa spinning disk scan heads (X1 and W1), each equipped with two cameras (iXon Ultra EMCCD and Neo CMOS). Aim of the GerBI Educational Programme is to foster the exchange of know-how and to advance the level of expertise and professional competence within the Bioimaging Community, in particular among managers of Imaging Core Facilities. The Job Shadowing Programme promotes visits of Core Facility staff members to other Core Facilities to learn more about the operation procedures of such facilities and their teaching activities and/or about a specific imaging technique. The One Place for GerBI Programme enables interested scientists to take part in a training Course provided by Core Facilities or Research Groups outside their own institution.
